遵义医学院学报ACTA ACADEMIAE MEDICINAE ZUNYI第32卷第5期2009年10月Vo1.32No.5Oct.2009___________________________[收稿日期]2009-03-14[基金项目]遵义医学院博士启动基金,贵州省国际合作项目基金(2009GZ52794),贵州省科学技术基金。[作者简介]陈阳(1977—),男,博士,副教授,研究方向:中药有效成分的提取,分离及分析。栀子为茜草科植物栀子Gardenia jasminoides Ellis 的干燥成熟果实,具有抗炎,镇痛和解热的功效。西红花苷和栀子苷是栀子药材发挥各种药理效果的物质基础,是这个药材质量评价的重要指标(结构见图1)。从栀子果实中分离提取栀子黄色素的方法有:浸提法、大孔吸附树脂法、超声波法、微波提取法、CO 2超临界提取法、无机膜提取法等。从栀子果实中分离提取栀子苷的传统方法有乙醇回流法和水煎法,近年来开发出了渗漉法、超声波提取法、大孔树脂吸附法等[1~5]。鉴于乙醇成本低,回收方便,毒性不大,国内药厂主要采用乙醇作为栀子化学成分提取溶剂,然而,至今国内未见报道对比研究不同浓度乙醇对于栀子中西红花苷和栀子苷提取效率的文献。本文采用不同浓度乙醇提取栀子化学成分,以西红花苷和栀子苷作为评价指标,运用高效液相色谱法(HPLC)对提取液进行含量测定,以期对栀子药材的粗提工艺提不同浓度乙醇提取栀子有效成分效果的研究陈阳1,赵璨2,张浩2,蔡乐2(1.遵义医学院珠海校区化学教研室,广东珠海,519041;2.四川大学华西药学院生药学教研室,四川成都,610041)[摘要]目的采用不同浓度乙醇提取栀子化学成分,以西红花苷和栀子苷作为评价指标,以期对栀子药材的粗提工艺提供一定参考。方法以西红花苷-1、西红花苷-2、西红花苷-3和栀子苷为对照品,采用HPLC 测定不同浓度乙醇提取栀子溶液中西红花苷和栀子苷的含量。结果乙醇浓度对栀子中西红花苷和栀子苷的提取效率影响较大,极高浓度和极低浓度乙醇都不利于这两类成分的提取,而中等浓度的乙醇可以较好的将两类成分提取出来。结论生产中应该根据提取目标产物的不同而选择相应浓度的乙醇,以利于使用最少的溶剂达到最大的提取效率。[关键词]栀子;西红花苷;栀子苷;提取;HPLC [中图分类号]R 284.2[文献标识码]A [文章编号]1000-2715(2009)05-0439-03Study on efficiency of different concentration of alcohol for extraction of biological chemicals from gardeniaCHEN Yang 1,ZHAO Can 2,ZHANG Hao 2,CAI Le 2(1.Department of Chemistry,Zunyi Medical College Zhuhai Campus,Guangdong Zhuhai 519041,China;2.West China School of Pharmacy of Sichuan University,Sichuan Chengdu 610041,China )[Abstract]Objective To investigate the relationship between geniposide,crocins contents and ethanol concentration used in the extraction of gardenia fruits in order to obtain data for crude extraction of gardenia .Methods Different extractions were obtained by various concentration of alcohol.Pure crocins and geniposide were used as standards for HPLC determination of samples.Results alcohol concentration significantly influences efficiency of extracting of crocins and geniposide from gardenia .Extreme high or low concentrations of alcohol seem negative for the extraction of these biological chemicals.Our data indicated moderate concentration of alcohol is beneficial for the extraction of crocins and geniposide from gardenia .Conclusions Concentrations of alcohol should be determined on the basis of aim chemicals from gardenia in order to obtain the best efficiency of solvents.[Key words]gardenia ;crocins ;geniposide ;extraction ;HPLC439··
